Caracterización fenotípica y molecular de variedades de caña de azúcar cultivadas en el estado de Tabasco

Abstract

Se llevó a cabo una caracterización morfológica de 12 variedades de caña de azúcar del estado de Tabasco mediante descriptores morfológicos, se identificó la filogenia por medio de secuenciamiento de la región ITS de DNA ribosómico y trnL-F cloroplástico, y se hizo una caracterización molecular usando la técnica AFLP. La descripción fenotípica se llevó a cabo considerando las características de entrenudo, hoja y yema. En la amplificación del DNA ribosómico se utilizaron los iniciadores ITS4 e ITS5. Para la amplificación del cloroplasto se utilizaron los iniciadores universales trnL-F. El procedimiento para la realización de los AFLPs se realizó siguiendo las recomendaciones descritas en el Kit AFLP®Analysis System I y AFLP® Starter Primer Kit. Entre las 12 variedades de caña de azúcar evaluadas se encontraron diferencias fenotípicas; en lo que respecta a la forma del entrenudo se formaron tres grupos diferentes. El grupo I caracterizado por su forma cilíndrica lo integraron las variedades, C 87-51, Mex 91-130, ATM 96-40, RD 75-11, Mex 79-431, SP 70-1284, CP 72-2086, Mex 69-290 y Mex 68-P-23. El grupo II caracterizado por su forma cóncavo-convexo lo integraron las variedades Mex 57-1285 y B 4362 y en el grupo III lo conformó la variedad Mex 59-32 quien presentó forma abarrilado. En la forma de la yema seis variedades presentaron ovoide, el resto presentó formas diferentes, en la conformación de la hoja predominó la forma erecta con el ápice curvado. La amplificación y secuenciación de la región trnL-F de cloroplasto y de la región ITS del DNA ribosómico, permitió la identificación de las variedades de caña de azúcar del estado de Tabasco encontrándose que pertenecen a Saccharum officinarum. En la evaluación molecular con la técnica AFLP, la combinación E-ACC/M-CTA fue la que mostró mayor polimorfismo, el tamaño de los fragmentos polimórficos osciló desde 72 hasta 1353 pb. El dendrograma reveló tres grupos distintos de Saccharum spp. El grupo I comprendió las variedades C 87-51, ATM 96-40, B 4362, Mex 69-290, Mex 57-1285 y Mex 91-130, las cuales presentaron un 0.77% de similitud genética. El grupo II comprendió las variedades, RD 75-11, Mex 79-431, SP 70-1284, Mex 59-32 y CP 72-2086, las cuales formaron un conglomerado, por consiguiente un 0.70% de características genéticas similares. El grupo III lo integró la variedad Mex 68-P-23. De las variedades analizadas esta última fue la que presentó una menor similitud genética con un 0.22% con el resto de las variedades._______A morphologic characterization of 12 varieties of sugarcane of from Tabasco state by using morphologic descriptors was performed, the phylogeny by sequencing of chloroplast region ITS of ribosome DNA and trnL-F was identified, and a molecular characterization was done by using technique AFLP. The phenotypic description considered the characteristics of internude, leaf and bud. In the amplification of the ribosome DNA ITS4 and ITS5 primers were used. For the amplification of the chloroplast the universal were used trnL-F initiators. The AFLPs were done following the described recommendations in the Kit AFLP®Analysis System I and AFLP® Starter First Kit. Among the 12 varieties of evaluated sugarcane had phenotypic differences; regarding internude form three different groups were formed. The varieties C 87-51, Mex 91-130, ATM 96-40, RD 75-11, Mex 79-431, SP 70-1284, CP 72-2086, Mex 69-290 and Mex 68-P-23, characterized by a cylindrical internude integrated. The second group integrated by the varieties Mex 57-1285 and B 4362 presented concave-convex internude the variety Mex 59-32 presented abarrilado internude. Regarding to bud six varieties presented ovoid form, the rest presented different forms. The standing the with the curved apex was the most common leaf. The amplification and sequencing of the region trnL-F of chloroplast and region ITS of the ribosome DNA, allowed the identification of the 12 varieties of sugarcane and showed that they belong to the Saccharum officinarum species. In the molecular evaluation with AFLP, the combination E-ACC/M-CTA showed the major polymorphism; the size of polymorphic fragments ranged from 72 to 1353 pb. The dendrogram revealed three different groups from Saccharum spp. The group first included the varieties C 87-51, ATM 96-40, B 4362, Mex 69-290, Mex 57-1285 and Mex 91-130, which presented a 0.77% of genetic similarity. The second group included the varieties RD 75-11, Mex 79-431, SP 70-1284, Mex 59-32 and CP 72-2086, which formed a conglomerate, with a 0.70% of genetic similarity. Group III range as part of the Mex-68 P-23. Of the evaluated varieties the latter presented a lower genetic similarity with a 0.22% compared to the varieties.
